Ernesto Bertarelli nació en Roma el 22 de septiembre de 1965. Sus padres fueron Fabio y María Iris Bertarelli.
Su padre trabajó para la compañía farmacéutica Serono, que luego compraría. La firma tomó fama internacional gracias al uso de la hormona de fertilidad Pergonal. El primer bebé probeta nacería gracias a Pergonal.
Ernesto estudió en la Universidad de Babson y se graduó en 1989. Luego asistió a Harvard Business School y obtuvo una Maestría en Administración en 1993. Tres años más tarde sería designado como CEO de la compañía Ares-Serono, que pertenecía a su padre.

En el 2006 las acciones de la familia Bertarelli fueron vendidas a Merck KgaA por la suma de $8,6 mil millones (€7,7 MM). Así nació Merck-Serono.
Cuando vendieron la compañía, Bertarelli y su hermana se enfocaron en la recientemente creada Fundación Bertarelli, dedicada a la conservación marina y la investigación científica.
Ernesto es un aficionado a los yates y en el 2000 creó el equipo Alinghi, que ganó la Copa Louis Vuitton en el 2003 y la Copa de América.
El éxito del equipo fue tan importante que el presidente de Francia Jacques Chirac premió a Bertarelli con el título Chevalier de la Legion d’honneur en el 2003. También mereció la distinción Cavaliere di Gran Croce del presidente italiano Carlo Azeglio Ciampi.
Ernesto cada vez se adentraba más en el mundo de la competencia de los yates. Entonces ordenó la construcción de un yate al astillero Appledore, que fue lanzado al mar en el 2008 con el nombre de Vava II. Se describió como el yate más grande, más largo y más sofisticado que jamás se había construido en Gran Bretaña. Este mega yate está valorado en $100 millones (€89,8 millones) tiene 96 metros de largo.
En mayo del 2013 Bertarelli creó el Campus Biotech, que se convirtió en el nuevo centro de la neurociencia en Suiza y que lleva a cabo importantes investigaciones que tienen repercusión mundial.
